Potentiometric and spectroscopic studies on the coordination of Cu(II) ion with aminophosphonic inhibitors of leucine aminopeptidase have shown that 1-amino-2-alkoxyalkylphosphonic acids and 1-amino-2-(N-alkylamino)alkylphosphonic acids are very effective chelating agents. Besides the α-amino group, the PO32− group is the major binding site for the metal ion, although its relatively low basicity excludes the possibility of monodentate coordination of this group, as was found for the simple aminophosphonates. The side-chain groups, and especially the substituted nitrogen donors, may compete with phosphonate oxygen in basic solution. The results obtained for the aminophosphonates are compared with those on their aminocarboxylate analogues.
